Well the Great San Francisco Earthquake of 1906 killed roughly 3,000 people (at the minimum) and left 300,000 people homeless (out of a population of about 410,000) (I'm getting these facts from Wikipedia) so I'm thinking anything in that range or above would be pretty devastating.

The population is much greater and more concentrated today, though, so many more will probably be killed. As for San Francisco....
